Research Abstract |
Invasion deficiency of the chorionic villi to the basement decidual layr is a well known pathology in preeclampsia (PE) and in some intrauterine growth retarded pregnancies. In the process of placental development, involvement of fibrinolytic factors, extracellular matrix (ECM) and immunologic system are suggested. The object of this study is to evaluate the participation of fibrinolytic factors and ECM possibly regulated by immunological system in the developing placenta. Study designs are ; 1) Comparison of fibrinolytic factors, ECM and cytokine levels in early 2nd trimester amniotic fluid between normal outcome and potential PE group, 2) Measurement of fibrinolytic factors and FFN in placental culturesupernatant with addition of cytokines. The results were ; 1) Significantly high levels of FFN and decreased PAI-C concentration in potential PE group compared to the normal outcome group of early 2nd trimester amniotic fluid. Among the several cytokines measured, IL6 and IL8 had significantly higher concentration in potential PE amniotic fluid, compared to the normal outcome group, and these two showed significant correlation. IL8 and FFE also had positive correlation. 2) In culture supernatant, FFN secretion by villous cells increased with addition of IL8 in a dose dependent line. 3) Both IL6 and IL8 concentrations were remarkably higher in decidual supernatant compared to villous supernatant. These results suggested the involment of fibrinolysis and ECM in the development of the placenta and the onset of PE,and a regulation by cytokines of maternal origin.
