Research Abstract

To clarify the mechanism of the recurrence of cytomegalovirus (CMV) retinitis, characteristics of human retinal pigment epithelial (RPE) cells to CMV infection were studied. The RPE cells could be infected with CMV.The result means that RPE cells may be one of the main targets of CMV infection. Pseudo-sensitivity of RPE cells to CMV was lower than that of human embrionic lung fibroblast (HEL) cells. The RPE cells were semi-permissive for virus replication because of quantitative or qualitative restriction of viral immediate early gene expression. CMV could recover from the CMV-inoculated RPE cells with no cytopathic effects (CPE). The RPE cells, therefore, appear to permit a latent or a small size of persistent infection with CMV.An anti-CMV effect of ganciclovir (GCV) was, in addition, compared between RPE and HEL cells, and more responsiveness of RPE cells to GCV was made clear. Therefore, not a sensitivity of RPE cells to CMV and a characteristic of RPE cells to GCV,but a latent or a small size of persistent infection of RPE cells with CMV may account for the hardness of CMV retinitis to be cured.
