| Research Abstract |
われわれは、endothelin receptor antagonist及びagonist(ET_Areceptor antagonist BQ-123及びET_Breceptor agonist BQ-3020、萬有製薬)を入手し、既報したendothelin-1(ET-1)による家兎網膜血管一過性閉塞モデル(Graefe's Arch Clin Exp Ophthalmol231:476-481,1993)の硝子体に投与してET-1の網膜血管収縮作用に対する影響を研究した。その結果ET-1の強力な網膜血管収縮作用には主にET_Areceptorが関与し、一過性網膜血管拡張作用にはET_Breceptorが関与している可能性を示した(Life sci53:pp.PL111-115,1993)。さらにわれわれは水素クリアランス法により家兎の網膜血管流量を測定し、ET-1による家兎網膜血管一過性閉 モデルでは、著しい網膜虚血時に脈路膜血流量が増加することを示した。すなわち、ET-1の硝子体内投与により眼内血流のダイナミックな再分配が起きることが考えられ、網脈路膜に高濃度に存在するとされるendothelin receptorsが眼内血流調節(自動調節機構)に何らかの役割を果たしている可能性があると考えられた(submitted in Graefe's Arch Clin Exp Ophthalmol)。また現在眼内血流調節に大きな影響を与える家兎眼圧に対するendothelin-1の作用についても検討中である。
