Research Abstract

Genetic characterization of Borrelia isolates from the tick, Ixodes ovatus, I.persulcatus and the rodent, Apodemus speciosus in Japan were compared to members of the three genospecies of Borrelia burgdorferi sensu lato ; B.burgdorferi sensu stricto, B.garinii and B.afzelii.DNA relatedness of the I.ovatus isolates with strain belonging to the three genospecies was 50 to 64%.These results suggested that the strains isolated from I.ovatus in Japan differ from the three genospecies and should be classified as a new genospecies of B.burgdorferi sensu lato.We propose that strains isolated from I.ovatus should be classifed as B.jeponica sp.nov.We developed PCR-RFLP method based on the RFLP of amplified products from OspA and OspB gene for determination of genospecies.The method is simple and faster than conventional DNA homology method.
Borrelia subcultured for one year in vitro lost or showed reduced expression of OspC in comparison with the low-passage strains.The low-passage strains efficiently infected outbred mice.In contrast, the incidence of infection with the high-passage strains was low.Isolates from the bladders of mice inoculated with the high-passage strains expressed large amounts of OspC in comparison with those originally inoculated.These results indicated that OspC expression is related to the infectivity of Lyme disease borrelia.
