Research Abstract |
The relationship between biological activities of streptococcal pyrogenic exotoxins (SPE) produced by Streptococcus pyogenes and periodontitis, between biological activities of mitogenic factor (MF) from oral streptococci and periodonttitis were studied. SPEs were purified, and ELISA for detection of these toxins was achieved. These SPEs proliferated to human periferal blood mononuclear cells (PBMC), and promoted to growth T cell bearing TCR Vbeta2. TNF activity and IL-2 production were detected in culture supernatant of PBMC treated with SPEs, and induction of mRNA expression of TNF-alpha, TNF-beta, IL-2 and IFNgamma was observed in PBMC.Furthermore, We detected MF from culture supernatant of Streptococcus anginosus, and partially purified the factor. The MF proliferated to human PBMC,induced production of IL-2 and TNF,and expression of the cytokines mRNA.These results suggest that SPEs or MF from S.pyogens or S.anginosus isolated from oral cavity can be related to periodontitis.
