Research Abstract

種々のグルタミン酸受容体アゴニストを神経科学の領域に導入してきた経験を基に、代謝調節型グルタミン酸受容体アゴニスト・アンタゴニストの開発を行ってきた。今までは代謝調節型グルタミン酸受容体の選択的アゴニストとして(1S,3R)-ACPDというあまり協力でないアゴニストしか存在しなかったが、本研究により、L-CCG-IやDCG-IVに加え、cis-およびtrans-MCG-Iなどの新しい代謝調節型グルタミン酸受容体アゴニストが見出された。グルタミン酸の官能基の立体配座を伸展型に固定した(1S,1′S,2′R,3′R)-および(2S,1′S,2′R,3′S)-2-(2-carboxv-3-methoxymethylcyclopropyl)glycine(cis-およびtrans-MCG-I)は、強力な代謝調節型グルタミン酸受容体アゴニストであることが判明した。trans-MCG-Iのアゴニストとしての効力は、ほぼDCG-IVに匹敵するが、高濃度でもNMDA受容体を活性化することはなく選択性が高いことが予想された。培養脊髄細胞でforskolinによるcyclicAMP産生を有意に抑制する。cis-MCG-Iは神経細胞を脱分極しないがキスカル酸適用後は、長時間にわたって脱分極を起こすようになる。この脱分極の程度はかなり顕著で、一種の長期増強現象とみなすことができる。キスカル酸適用後に起こるcis-MCG-Iによる脱分極は、NBQXにより著しく抑制されるが、代謝調節型受容体アンタゴニストのMCPGでは抑制されない。神経情報伝達の生理機能を解明する有力なアゴニストはこの領域の研究に必要不可欠のものである。
