Research Abstract

収縮環形成のシグナル伝達系の因子の候補、低分子量Gタンパク質Rhoに関しては、分裂酵母Rho1が隔壁部に局在すること、Rho2の過剰発現が細胞形状変化や細胞壁の厚化を引き起こすこと、rho3、rho4遺伝子の破壊は細胞極性の異常や分裂異常を引き起こすことを見いだした。もう1つの候補であるミオシン軽鎖のリン酸化については、まずHeLa細胞から3種類のミオシン軽鎖遺伝子を単離した。次にリン酸化ミオシン軽鎖を特異的に認識する抗体を作製し、分裂期のHeLa細胞を染色したところ収縮環が染色された。またミオシン軽鎖のMLCK部位をリン酸化するキナーゼとしてMAPKAK2を同定した。ウニ卵の染色体分離と細胞質シグナル伝達の時間的関係をコルヒチンの顕微注入により調べた。テトラヒメナのEF1_αとF-アクチン結合タンパク質p60が分裂溝に存在することを見い出した。またテトラヒメナの分裂面決定に関与するp85の遺伝子の全塩基配列を決定した。ウニ卵の分裂溝に局在するアクチン調節タンパク質ABP40がアクチン繊維切断活性を持つことを明らかにした。さらにウニ卵の新規のF-アクチン結合タンパク質として見い出したp60とp150が分裂溝に濃縮されることを見た。またアフリカツメガエル卵の分裂に必須なアクチン調節タンパク質XACに結合する5種のタンパク質を見い出し、その1つがWDリピートを持つことを明らかにした。アフリカツメガエル卵のCa放出がIP3を介して行われることを示した。アクチン繊維を細胞膜に結合せさると考えられるERMファミリータンパク質は細胞膜タンパク質の塩基性に富む領域に結合することをICAM-2,IL-2についても確かめた。クラミドモナスのアクチン変異株を単離した。この株は細胞質分裂を正常に行った。
