Research Abstract

収縮環形成のシグナル伝達系因子の候補、低分子量Gタンパク質Rhoに関して、分裂酵母Rho2-4がRho1と同様に隔壁部に局在すること、Rho1とRho2は似た機能を持っているが重複はしていないことを見た。出芽酵母の出芽を制御するRHO3の欠損を多コピーで抑圧するSRO7がミオシン(Myo1,Myo2)と相互作用することを見い出した。RHO1の標的タンパク質であるBNI1はプロフィリン、EF1α、SPA2と結合することを示した。これらのタンパク質を介してアクチンに作用する可能性がある。哺乳類のRhoAの標的タンパク質としてp140mDia、p160ROCK、citronkinaseを見い出した。p140mDiaはプロフィリンを結合し、分裂溝に局在する。citronkinaseも分裂溝に局在し細胞質分裂に働いていることが知られた。分裂酵母の収縮環がアクチン繊維でできており、トロポミオシンを含むことを免疫電顕で確認した。分裂酵母から新しいミオシン重鎖の遺伝子myo3^+をクローニングし、Myo3がMyo2と協同的に収縮環の形成に働くことを示した。細胞性粘菌の分裂変異遺伝子を同定したところIQGAPにホモロジーのあるタンパク質だった(GAPA)。GAPAと同じファミリーのRasGAPの遺伝子を同時に破壊すると、細胞質分裂に強い影響が出たのでこれらのタンパク質が細胞質分裂に働いていることが分かった。ウニ卵の分裂溝に濃縮される新規のF-アクチン結合タンパク質として見い出したp60とp150がそれぞれミオシンVIとコロニンであることを明かにした。マウスのセプチンNedd5が分裂溝に局在し、その抗体のマイクロインジェクションが細胞質分裂を阻止することを見た。またNedd5の繊維形成にはそのGTPase活性が必要であることが分かった。カエル卵の分裂に必須なアクチン調節タンパク質XACに結合するタンパク質の一つAIP1と同定し、これがXACと協調してアクチンを脱重合することを見い出した。これらの結果を総括するため、海外、国内の専門家を招聘して2日間の国際会議を行った。
