Research Abstract

1.Ahリセプター(AhR)遺伝子を欠失したマウスを作成し、2, 3, 7, 8-四塩化ダイオキシン(TCDD)の奇形誘発作用を調べた。12.5日胚をもつ妊娠マウスに、経口投与によってTCDDを与え、18.5日胚を調べたところ、正常マウスでは全胎仔で異常が見られた口蓋裂と水腎症が、ノックアウトマウスでは全く生じなかった。
2.ArntはC末端部に転写活性化領域をもち、この活性がアデノウイルスの初期遺伝子ElAによって阻害されることはすでに報告したが、この転写活性化を仲介するコファクターとしてCBPとP300を同定した。
3.AhRと構造類似性をもつ抑制型転写因子をクローニングした。Arntとのヘテロダイマー形成活性、XREへの結合活性はAhRと同程度であり、AhRと競争的にXREからの転写を阻害することを見いだした。組織発現分布を調べたところ、多くの組織で発現しており、特に心臓での発現が強かった。
4.PASドメインをもち、HIF-1αと構造類似性をもつ正の転写因子HLFをクローニングした。Arntとのヘテロダイマー形成活性、DNAとの結合活性はHIF-1αと同程度であった。またHIF-1αと同じDNA結合特異性を示した。肺での発生過程での発現を調べた結果、HIF-1αによって転写が活性化されるVascular Endotherial Growth Factorの発現とよく相関しており、その転写活性化に関与していることが強く示唆された。
