Research Abstract

ヒトのがんの90%は上皮細胞に由来するカルシノーマである。われわれは上皮細胞のがん研究における重要性に着目して、その組織構築と増殖・分化のメカニズムをケラチノサイトをモデルとして、偏平上皮分化のシグナル伝達経路、上皮組織構築における増殖因子、発生がん過程における形態形成遺伝子の発現、偏平上皮分化に関する遺伝子操作動物の作成などを目的として研究を進めてきた。本年度は次の3研究項目について研究を行った。1)偏平上皮のシグナル伝達経路:上皮細胞に特異的に発現しているCキナーゼη分子種のトランスジェニックマウスを作成した。トランジェニックマウスは皮膚の角化亢進と代償性の表皮過形成を示した。2)細胞周期にともなうCキナーゼ各分子種の発現をBALB/MK-2細胞の同調培養系を用いて検討した。α、δ、ζ分子種は細胞周期の進行によっても発現が変わらなかったのに対し、η分子種はG1/S期に発現した。3)皮膚発がんにおけるHox遺伝子群の発現を検討した。パピローマ、カルシノーマではA7,B7遺伝子が特に強く発現した。
