Research Abstract

ツメガエル卵成熟と細胞がん化におけるMosの機能経路に関し、次のような成果を得た。
1.ツメガエル卵成熟を負に制御するプロテインホスファターゼ2Aが、Mosによる卵成熟開始に対しても負に働くことを明らかにした。
2.これまでの他のグループの報告と異なり、Mosの卵細胞分裂抑制因子(CSF)としての活性のためにcdk2キナーゼは不要であることを示した。
3.MAPキナーゼファミリーの特異的阻害タンパクであるCL100を用いて、Mosの下流に(古典的MAPキナーゼ以外の)MAPキナーゼ関連経路が存在することを示唆した。
4.NIH3T3細胞のがん化において、Mosの下流でMAPキナーゼがc-Fosの他にFra-1やFra-2もリン酸化し、かつ安定化させることを明らかにした。
5.しかしながら、c-Fosの場合と異なり、リン酸化・安定化されたFra-1,Fra-2のみではNIH3T3細胞をがん化できないことを示した。
