Research Abstract

B細胞抗原レセプターの分子Ig-α(MB-1)に結合する分子p52がB細胞活性化に伴って著しくリン酸化される。19-14抗体を用いて単離したcDNAクローン化α4には、典型的なシグナル伝達分子との相同性が無く、明らかなキナーゼドメインも存在しない。Ser/Thr,Tyrのリン酸化可能配列は多く、これらはin vitroのリン酸化試験でリン酸化された。またATP結合配列と、SH3結合部分様配列が認めれた。酵母菌の栄養刺激に必須なシグナル伝達分子(TAP42)がα4ホモローグであり、ラパマイシン経路に関わり細胞の生存、増殖、細胞周期回転に必須な分子であると考えられた。マウスα4をJurkatに遺伝子導入し、α4がリンパ細胞のサバイバルに関連するかどうかを調べたところラパマイシン感受性が解除された。マウスα4が酵母菌と同様なラパマイシン感受性経路に関わる分子であるかどうかを調べ、α4がphosphatase 2A(PP2A)と特異的に結合し、その結合はラパマイシン感受性であることを確認した。PP2Aは、cyclin B/P34^<odc2> protein kinase活性の制御に関与することが示唆されている。α4/PP2A関連シグナル伝達が細胞周期回転や細胞の維持に重要な機能を有していると予想された。またラパマイシン感受性とリンパ細胞の増殖、α4とphosphatase 2A(PP2A)との特性的結合に関する解析を行った結果、α4がリンパ細胞の増殖、細胞周期進行に密接に関連していることを見いだした。α4分子は酵母からヒトに至るストレス関連の分子群を構成している可能性が考えられた。このα4関連シグナル伝達経路の解析を進めることによって、細胞の生存維持の刺激伝達機構を明らかにするものと期待される。
