Research Abstract

1.酵母のtwo-hybrid遺伝子発現系を用いた癌遺伝子機能阻害物質の探索
Ras/RaおよびRas/CDC25相互作用阻害物質の検出系を構築し、これを用いて広範なスクリーニングを行ったが、現在までのところ新規の活性物質は得られていない。しかし、我々がすでにSrcキナーゼを阻害する細胞周期阻害剤として同定したラディシコールがこの系でのRas/Raf結合を特異的に阻害することが判明し、その作用機構の解析を行った。v-Ha-rasトランスフォーム細胞を用いて細胞内でのRas/Rafの結合に対する効果を調べたところ、それを強く阻害することが判明した。本物質は、in vivoでRas/Raf結合を阻害する初めての物質であると考えられる。一方、in vitroでの直接的な結合阻害は認められず、in vivoでの結合阻害には、Rafのキナーゼドメインの存在が必要であることが分かった。
2.Preussin耐性遺伝子のクローン化
我々は分裂酵母cdc25変異株に対し選択的に作用する物質としてPreussinを得た。その作用機構を解析する目的でcdc25変異株を用いその超感受性を相補する遺伝子のクローン化を行いHSP70をコードするhsp1/sks2をクローン化した。本遺伝子の破壊株は、生育可能であったが、増殖速度が低下し、破壊株とcdc25との2重変異株は、著しい増殖速度の低下、制限温度の低下、Preussin処理したcdc25変異株と同様の形態異常を示し、Preussinの作用はHSP70と関連する可能性が示唆された。
