Research Abstract

原がん遺伝子N-myc、E2-box結合因子δEF1などの転写制御因子の作用を研究した。
N-myc欠損マウス胚と正常胚との間のcDNAサブトラクションによって単離されたN-mycの制御標的遺伝子Ndr1についてN-myc蛋白質の作用を詳細に解析した。Ndr1遺伝子は上流10kbに位置するエンハンサーと、TATA-boxを持つプロモーターによって調節されていることが明らかになった。。N-myc:Maxヘテロダイマーがプロモーターに結合することによって、Ndr1遺伝子転写が抑制される。既知のc-mycの制御標的遺伝子がc-myc:Maxによって転写を活性化されることと対照的である。Ndr1は、動植物界に広く保存される細胞質の蛋白質であり、基本的な生理過程に関与していると推定される。N-mycの制御標的遺伝子が明かになったので、個々のMycに固有の制御機能を解析できるようになった。
δEF1は、Znフィンガーとホメオドメインを持つ転写制御因子である。さまざまのエンハンサー中のE2-box配列にZn-フィンガーを介して結合して、転写活性化因子をE2-boxから排除するとともに、N末端側の転写抑制ドメインによって、エンハンサー全体を抑制することを明かにした。δEF1欠損マウスは2つの異なった組織に欠陥を生じる。(1)T細胞分化の異常。T細胞前駆体がほとんど胸腺に見いだせない。この表現型は、Null突然変異体のみならずにC末端Znフィンガーの欠失のみで生ずる。(2)Null突然変異体に見られる骨格形成の異常。表現型の解析などから、δEF1がBMP、GDFなどのリガンドによって活性化される受容体の下流に位置することが示唆された。
