Research Abstract

HPVE6E7遺伝子によるin vivo発がんの解析
Leydig細胞腫瘍好発系E6E7Tgマウス(HPVE6E7トランスジェニックマウス)を用い、腫瘍発生に対するp53遺伝子の役割につき検討し、HPVのE6E7遺伝子による発がんにはp53の不活性化に依存する系(例Tリンパ腫)と非依存性の系(例Leydig腫瘍)が存在する可能性を示唆する結果を得た。
Transformation抑制遺伝子の検索
1.ラット初代培養細胞(REF)中に存在が予想されるHPVE6E7遺伝子やv-srcなどによるtransformationを抑制する遺伝子の分離をサブトラクション法により試み、REF細胞で特異的に発現している21種類の遺伝子について検討し、12種類が未知遺伝子、残り9種類が既知遺伝子であることが分かった。上記既知遺伝子中、性状解析を行ったlumican遺伝子はH-ras遺伝子によるtransformationを抑制する結果を得た。
2.proteoglycanの一種であるsyndecan-1のm-RNA発現が調べた子宮頚癌由来のすべての細胞株で特異的に減少しており、syndecan-1遺伝子を過剰発現させると細胞増殖能や、軟寒天培地中でのコロニー形成能を抑制させることを見い出した。
3.v-srcやv-K-rasなどのがん遺伝子の発現によってdown regulateされる新しい遺伝子(drs)はこれらがん遺伝子によるtransformationに抑制的に働くことを明らかにした。
