Research Abstract

(1).無治療の非小細胞肺癌232名についてCYP1A1,GSTM1のgerm lineの遺伝子多型と各種臨床的パラメーター及び予後との関係を検討した。喫煙由来の肺癌に対して高感受性のCYP1A1(B+C型),GSTM1(-)を持つ患者においては進行例が有意に多く,予後不良因子との相関が観察された。予後因子としてのCYP1A1多型を検討するためにステージIIIa-IV期の非切除非小細胞肺癌98例について主な予後因子別に遺伝子型による生存期間を検討したところ,全ての因子においてB+C型の方がA型よりも有意に予後不良であることが明らかになった。Coxの比例ハザードモデルでの多変量解析の結果,CYP1A1多型はPSの次に強い独立した予後因子であった(HR=1.98;95%CI 1.24-3.17;p=0.005)。
(2).CYP1A1の誘導的発現に関与する転写因子Ahr (Aryl hydrocarbon receptor), Arnt (Ahr nuclear translocator)の核局在性のメカニズムを解析した。β-ガラクトシダーゼとの融合タンパクの細胞内局在性,及びGST-GFPとの融合タンパクのmicroinjectionの結果より,Arntは核タンパクであることが明らかになった。NLSとしてArnt39-61が同定され,塩基性及び酸性アミノ酸のクラスターで構成されているという興味のある結果が得られた。一方,Ahrもβ-ガラクトシダーゼとの融合タンパクの局在性の解析によりAhr12-18の塩基性アミノ酸クラスターがNLSのコアとして機能する事が明らかになり,Ahrのリガンド依存性核移行はHsp90の結合によりNLSがマスクされており,リガンドの結合によりHsp90が解離し,それに伴う構造変化によることが示唆された。
