Research Abstract

For the purpose of promoting tissue (bone, tendon, peripheral nerve) allograft as a common clinical work, we studied the method of 1) the most suitable treatment of the tissues for allograft, and 2) safer management of the grafting tissues, and then 3) on the basis of the results we establish a tissue bank in both our hospital and regional area.
1) Tissues (bone, tendon, peripheral nerve), which were harvested from BN rats, were preserved as three groups ; (1) rapid freez materials of -80゚C in a freezer, (2)slowly stepped freez material of -80゚C in a programing freezer, and (3) immediately after warming, rapid freez materials of -80゚C.Three months later, the materials of each group were grafted in Lewis rats. Every month, bone union was checked radiographically, and the grafted tissues were observed histopathologically. The results, which were almost similar in the three groups, means that the most simple way is rapid freez in -80゚C freezer in order to reduce antigenecity of the grated tissues.
2) For confirming a safer management of the tissues, the harvested tissues from BN rats were preserved in -80゚C freezer as three groups ; (1) double package in polyethylene bags, (2) triple package in polyethylene bags, (3) kept in a small bottle with packing by the bags. Six months later, the tissues were grafted into Lewis rats with bacteriological investigation. The results showed that no bacteia was proved in any samples, and no pyogenic infection or other immunological problems occured in the grafted tissues.
3) For establishing tissue bank, we made a standard method for tissue allograft in our hospital, and the tissue meterials were provided to other hospitals in a regional areafor thier tissue allografting. We would make a further co-ordinate system in the regional hospitals.
