Research Abstract |
1) We measured the cell envelope structures of Gram negative anaerobic bacteria, Prevotella intermedia ATCC 25611 and ATCC 33563, and Porphyromonas gingivalis 381 after shade processing and threshold setting using the LUZEX-FS image processor. Bacterial samples were fixed by glutaraldehyde-OsO_4 (GA-OSO_4) and freeze substitution using OsO4-acetone (FS). The width of cell envelope (between inner leaflet of cell membrane and outer leaflet of outer membrane) of 25611 strain was 39.3 nm by GA-OsO_4 and 31.5 nm by FS,and those of 381 strain were 35.8,37.0 and 35.8 nm, respectively. The width of each aspect of the 25611 strain cell envelope by GA-OsO_4 fixation (FS fixation) was 13.0 (11.9) nm for the cell membrane, 4.6 (3.8) nm for the inner leaflet, 2.7 (4.4) nm between the inner and the outer leaflets, 3.6 (3.3) nm for the outer leaflet, 6.2 nm for the peptidoglycan layr, 9.2 (11.0) nm for the outer membrane. f the cell membrane and the inner leaflet of the outer membrane) of bacteria by GA-OsO_4 was larger than that beside outer membrane. 2) We studied the ultrastructure of bacteria in host cells to understand the appearance of oral infectious diseases using the image processor. Electron microscopic samples were taken of pus from patients with abscesses. Gram positive and negative bacteria were detected in almost all the abscesses used after anaerobic culture and electron microscopic observation. The area-ratio of bacteria to phagosomes was 33.0 % for gram negative, and 43.8 % for Gram positive bacteria. The area-ratio of the capsule to bacteria was 33.0 % for Gram negative, and 26.7 % for Gram positive bacteria. Vesicles originating from the outer membrane of Gram negative bacteria were about 20.6 nm in diameter and had an area of 541 nm ^2. The total lysosome area was about 20.5 % of the host cell cross section area, while this area was 21.1 % foe the phagosome.
