Research Abstract |
To investigate the roles of osteonectin (SPARC) in arthritis, we deter mined the level of osteonectin in cartilage and synovium specimens and synovial fluids from rheumatoid arthritis (RA) and osteoarthritis (OA) patients. Osteonectin in these tissues was immunostained. Osteonectin synthesis by chondrocytes in culture was determined by Northern blot analysis, immunoblotting, and sandwich enzyme-linked immunosorbent assay. The results showed that osteonectin synthesis in RA and OA synovium and cartilage is markedly enhanced. TGF-b and BMP-2 increased osteonectin synthesis by cultured chondrocytes. The osteonectin level in synovial fluids was 10-fold higher in RA than in the OA population. These findings suggest that various growth factors and cytokines regulate the production of osteonectin by chondrocytes at pre- and posttranlational levels, and that osteonectin is useful as a marker for arthritis.
