Research Abstract |
1.Extraction and analysis of worm protein Antigenic proteins were extracted by sonication or freezing and thawing from first- and second-larvae and also from adult worms, and analyzed by SDS-PAGE.Twenty-five to thirty protein bands of 28-220 Kda were detected. Western blotting assay showed that 56 and 200 Kda proteins were specifically reacted with artificiallyinfected rat sera. 2.Serological diagnosis for angiostrongylosis ELISA and latex aggluti-nation test (LA) were compared. Non-specific reactions were seen when hemorrhagic wild rat sera were tested with ELISA but not with LA.Thus LA was applied. Latex particles (1.053 in diameter) were coated with antigenic protein (1.78 ug/ml). Good correlation between ELISA and LA was obtained. The cut-off value was determined as serum dilution of 1 : 400 based on the bimodal distribution of antibody titers of 638 wild rats. Positive rate was 25.5% (163/638) of wild rats in Nagoya city. By cont-rast, 53.3% (8/15) of rats in Okinawa was seropositive. More research is needed for extraction of specific antigen from worms but LA is seemed to be a usefu1 diagnostic tool of angiostrongylosis.
