Research Abstract |
Esophageal dysplasia has been considered as atypical epithelial lesions. We investigated 12 lesions of the esophageal dysplasia for characterization on immunohitochemisty and mutation analysis of p53 gene. All lesions were picked up by fine needle and deparaffinized for PCR.PCR products were analyzed by direct sequencing method on auto-DNA sequencer. Some samples were analyzed after subcloning and SSCP.Immunohitochemistry showed positive staining of the nuclei in more than half of the dysplastic lesions. All of the severe dysplasia showed marked positive staining. Five of 10 lesions in the moderated and severe dysplasia also had mutations of p53 gene. 1 lesion showed missense mutation at codon 175, and 3 lesions at colon 273. One moderate dysplasia showed nonsense mutation at condon 176. None of the mild dysplasia showed p53 gene mutation. Morphological examination revealed precancereous lesions, but our results of p53 gene analyzes showed that esophageal dysplasia could not be distinguished froma carcinoma of the p53 gene level. Then, the esophageal dysplasia may be very close to intraepithelial carcinoma.
