Research Abstract |
From previous experiments, it was expected that an unknown bone marrow stromal cell derived myeloma cell growth factor playd an important role in the growth of myeloma cells. We have analyzed the growth factor activity to understand the mechanisms of myeloma cell growth. Following data were obtained : 1) Three bone marrow stromal cell dependent myeloma cell lines, ILKM10, ILKM12 and ILKM13, were established. 2) All three cell lines grew absolutely dependently on bone marrow stromal cells. They did not proliferate in response to known cytokines including IL-6 or gp-130-related cytokines, but proliferate in response to bone marrow stromal cell culture supernatants. 3) The growth factor activity included in the culture supernatant was semipurified utilizing an ion exchange column chromatography and an anti-IL6 antiboy affinity column, and the myeloma growth factor activity was separated from IL-6. Data confirmed that at least another unknown factor playd an important role in the growth of myeloma cells.
