Research Abstract |
The development of molecular genetics has progressed the concept of gene therapy from the theoretical to reality. The current wave of gene therapy will suggest the application of gene therapy to not only genetic disorders but also acquired diseases, such as cancer or infectious diseases. The purpose of this study is to investigate the role of tumor necrosis factor receptors (TNF-RI and TNF-RII) as one of therapeutic gene for lung cancer gene therapy. First, we analyzed the mRNA expression of human TNF-RI and TNF-RII in primary lung cancer specimens and normal lung tissues from cancer patients. The results demonstrated that lung cancer expresses TNF-RI mRNA and normal lung tissue expresses both TNF-RI and TNF-RII mRNA simultaneously. Further, TNF-mediated tumor cell lysis was observed in all lung carcinoma cell lines that exhibited the TNF-RI mRNA.Therefore, lung cancer cells expressing at least TNF-RI may be responsive to TNF therapy. On the basis of these findings, we constructed the two expression vectors of TNF-RI and TNF-RII cDNA.These vectors would be useful for further investigation of tumor necrosis factor receptors gene transfer into human lung cancer.
