Research Abstract |
The invastigators arranged a photo-multi-intensifier, a photo-counter, a light guide and a dark-box to make up a chemiluminescent measuring system in 1994. The system was revealed to have a capability of measuring the chemiluminescence using MCLA as a increasing factor for the chemiluminescence in the rat's air pouch model in the dark-box. The MCLA is very sensitive material, and its concentration effects much to the singularity of detecting the superoxide. The pourpose of the study during 1995, is to establish the adequate methods of the measurement. The invastigators made a air pouch model on the back of the rat at age of 6 to 8 weeks, the body weight of which were 200 to 240 g. Picnin-A and Corynebacterium parvum were used as a stimulant for the leukocytes in the air pourch. Two ml of Picnin-A and Corynebacterium parvum solution were injected into 7-days-old air pouch. The concentrations of the Picnin-A and Corynebacterium parvum solution were 0.1%, 0.5% and 1.0%. At 3,6,12,24 and 48 hours after injecting a Picnin-A solution, and at 48 hours after injecting a Corynebacterium parvum solution, the chemiluminescences were measured using 2 ml of 20mus of MCLA.The chemiluminescence incresed as the consentration of Picnin-A and Corynebacterium parvum increased. The kind of stimulant influenced the amount of chemilumineses. The duration between the injection of the stimulant and measurement did not effect the amount of chemiluminescence.
