Research Abstract

I examined influence of radiation, coaction of steroid hormone, thyroid hormone and radiation for human skin cultivation fibroblasts.
Method : I did culture passage of human skin cultivation fibroblast, and used alpha cell of passage early stage for simulation. The radiation exposure could control dose precisely, and, in wire form, I selected^<60> Co gamma-ray by it ising possible for constant, and did reproducibllity with 0.3Gy/min, 5Gy.10Gy.20Gy using telecobalt apparatus. Because reproducibility of dose wasn't fixed in other wire form, I didn't use it for simulation. I measured alpha cell number after 72 hours and, after exposure right after, 24 hours, evaluated proliferation of cells ability to examine gamma-ray exposure influence, and observed alpha cell morphology by light microscope and scanning type/transmission electron microscope in exposure right after, after 24 hours.
Next, 0.1,1.0,10.0 mu g/ml each added Thyroxine (T4) and Hydrocortison (HC) in culture medium, and 0.7Gy/min, … More 10Gy irradiated gamma-ray using telecobalt apparatus, and I measured alpha cell number after exposure right after, 24.48 time and evaluated proliferation of cells ability, and observed alpha cell morphology by transmission electron microscope in after 24 hours.
Consequence : It was accepted an increase of human being cutis cultivation fibroblast to be restrained by gamma-ray exposure. About alpha cell morhology, the differential wasn't found with light microscope, but smoothing of decrement and cell surface of microvilli was looked at with gain of dose with scanning electron microscope. And, in transmission electron microscope, alteration of morphology of gain and mitochondria of residual body was looked at, and, in particular, was looked at with low dose by after 24 hours, and residual body remained with high dose even after 24 hours.
That growth of human being cutis cultivation fibroblast was restrained was recognized by adding T4, HC in culture medium than I irradiated a merely gamma-ray in HC too. And, there was the determination which promoted proliferation of cells in T4, and it was thought whether you didn't reduce barrier by radiation exposure.
Problems : Some differences are recognized more by experimental findings to boil it, but correspond to a difference of radiation sensitivity of individual cultured cell by using the same year charges, the same chromosome region for primary culture. Proliferation of cells ability is restrained by radiation exposure, but because the alteration which was wasn't recognized in observation between short terms, I think about alpha cell morphology when want to observe a long term than the future, and think when want to do analysis by gene level more too.
