Research Abstract |
Recent study have characterized and cloned two type of cyclooxygenase. Cyclooxygenase type 1 (COX-1) is constitutive form. On the other hand, cyclooxygenase type 2 (COX-2), a new form of cyclooxygenase, is expressed only after stimulation by various cytokines. Cells (amnion-derived WISH cells) were cultured and stimulated by phorbol myristate acetate (PMA). The PGE2 in culture media were measured by ELISA.Total RNA was extracted and analyzed with Northern blotting analysis. In the time course of PGE2 production in the presence or absence PMA,the concentration of PGE2 was not detected until 2h of incubation, but was detected after 4h. The stimulation and suppression of PGE2 production in the presence or absence PMA and/or DEX,PGE2 was significantly increased by PMA and suppressed by PMA and DEX,and DEX only. In the time course of COX-2 mRNA in the presence or absence PMA,the band of COX-2 mRNA was detected after 1h. The most strong band of COX-2 mRNA was detected at 2h. In the stimulation and suppression of COX-2 mRNA in the presence or absence PMA and/or DEX,the band of COX-2 mRNA stimulated by PMA was strongly detected and the band stimulated by DEX was not detected. These results suggested that COX-2 was present and expressd by activated protein kinase C.The COX-2 might be important role of labor and deliverly in parturition.
