Research Abstract

Three days after the cryotreatment on mouse lip skin, epidermis was detachd from the basement membrane, forming a blister cavity. The epidermal basement membrane was thick in some parts. These thick parts were thought as lamina densae below hemiesmosomes. Regenerating epidermal cells migrated on the basement membrane from hair follicles in the form of multiple cell layrs connected by desmosomes. By the immunohistochemistry on the ultrathin sections for transmission electron microscopy using anti-heparan sulfate proteoglycan antibody and colloidal gold conjugated secondary antibody, positive reactions were observed on the thick parts of the epidermal basement membrane. Regenerating epidermal cells reconnected with the basement membrane, forming hemidesmosomes, and the thick parts of the basement membrane below hemidesmosomes showed the HSPG-positive reactions. It was stongly suggested that regenerating epidermal cells make contacts with the basement membrane at the places of old hemidesmosomes.
Regenerating epidermal cells did not closely connect with the basement membrane, but there was some space between the cells and he basement membrane, showing there were electron-lucent material in lamina lucida. Observations by ultracryomicrotomy and negative staining showed the thicker lamina densa and thinner lamina lucida than in the conventional electron microscopy. This fact supports the existance of electron lucent substance in lamina lucida.
