Research Abstract |
ABNORMALLY MIGRATING ALKALINE PHOSPHATASES (AP_2) ARE DETECTED MORE FREQUENTRY BY POLYACRYLAMIDE GEL DISC ELECTROPHORESIS (PAGE) THAN WITH CELLULOSE ACETATE MEMBRANE,A HIGH-MOLECULAR WEIGHT INTESTINAL AP (HIAP) DETECTED IN HEALTHY SERUM AND ASSOCIATED WITH THE ABH AND LEWIS BLOOD GROUPS WAS INVESTIGATED.BY PREINCUBATION OF BLOOD GROUP B OR O SERA AND A MONOCLONAL ANTIBODY TO HUMAN INTESTINAL AP,THE HIAP WAS FOUND AT THE ORIGIN OF PAGE.IT WAS ABLE TO NORMALIZE HIAP BY BRIEF TREATMENT WITH SUBTILISIN OR PAPAIN.THE RESULTING AP WAS EXHIBITED TO DETECT THE SAME MOBILITY AS COMMON INTESTINAL AP.BLOOD APPEARANCE OF HIAP WAS SPECIFICALLY DEPENDED ON HEALTHY BLOOD GROUP O AND B SUBJECTS BEARING WITH LEWIS b BLOOD GROUP (SECRETARY TYPE). WHEN THE TESTED SUBJECTS BEARING LEWIS a BLOOD GROUP (NON-SECRETARY TYPE) WAS USED,THERE IS LITTLE IF ANY DETECTED THE HIAP IN THEIR CIRCULATIONS. BASED ON THE RESULTS OF GEL FILTRATION,THE MOLECULAR SIZE OF THE GLYCOPROTEIN BINDING TO INTESTINAL AP HAD APPROXIMATELY 30 KILODALTONS WITHOUT THE INTESTINAL AP AND A DETERGENT.HOWEVER,THE GLYCOPROTEIN BOUND TO INTESTINAL AP IN THE PRESENCE OF TRITONX-100 WAS FOUND AT THE SAME MOBILITY AS THE HIAP IN BLOOD GROUP O AND B SUBJECTS BEARING LEWIS b BLOOD GROUP.FURTHERMORE,IT IS INTERESTING THAT CIRCULATING AMOUNTS OF THE GLYCOPROTEIN BINDING TO INTESTINAL AP ARE MATCHED APPARENTLY TO THE BLOOD GROUP B OR O-TRANSFERASE. TAKEN TOGETHER,THE BLOOD APPEARRANCE OF INTESTINAL AP IN HUMAN SUBJECTS MAY BE RELATED STRONGLY TO THE BLOOD GROUPS ABH AND LEWIS.
