Research Abstract |
In order to obtain a clue to estimate whether brain cholesterol is synthesized in situ or not, occurrence of a P450 enzyme catalyzing lanosterol 14-demethylation (P450_<14DM>) was examined. Incubation of lanosterol with rat brain microsomes under the presence of NADPH and molecular oxygen formed the known 14-demethylated metabolite. This metabolism was inhibited by ketoconazole, a potent P450_<14DM> inhibitor as well as CO.These facts strongly indicated the occurrence of P450_<14DM> in rat brain microsomes. The brain demethylase activity was altered a little during maturation. In situ hybridyzation experiment with rat P450_<14DM> cDNA revealed that P450_<14DM> mRNA was expressed at the limited site of the brain. Lines of evidence obtained in this study suggested the occurrence of de novo cholesterol biosynthesis in brain.
