Research Abstract |
The ras-related small GTP-binding proteins have been implicated in the regulation of various cellular processes, ranging from growth, differentiation and cytoskeletal organization to intracellular vesicle transport and secretion. In plants, genes encoding these proteins have also recently been identified, but little is known of the mechanisms that regulate the genes or of the functions of their encoded proteins. We have isolated one such gene from wheat, wgp7, which shows a rapid (within 30 min) and dramatic (20-fold) increase in transcript levels in response to treatment with sulphur dioxide (SO_2) or nitrogen dioxide (NO_2), but not to a range of similar compounds. Further analysis has shown that the gene is also induced by factors that increase the level of free oxygen radicals, suggesting that wgp7 may be involved in plant responses to oxidative stress. Transgenic plants expressing wgp7 have been constructed and expression of the gene confirmed. Experiments testing the responses of these plants to various oxidative stresses, and to SO_2 and NO_2, are underway. Furthermore, transgenic plants containing pwgp7-GUS fusions are being constructed to identify regions in the promoter of wgp7 which provide SO_2/NO_2 inducibility. Recombinant wgp7 proteins are being synthesized and will be used to raise antibodies for localizing wgp7. (和訳) ras関連のGTP結合タンパク質は細胞の増殖、分化、細胞骨格形成、膜糸への転送、分泌等、多面的な機能に関与する。 近年、これらのタンパクをコードする遺伝子が検出されているが遺伝子の発現制御、産物の機能については不知な部分が多い。我々は、このような遺伝子の一つWgp7を小麦より得た。この遺伝子の転写物は二酸化イオウ、二酸化窒素添加によりすみやかに(30分以内)、かつ、大量に蓄積することを見いだした。さらに当遺伝子は酸素ラジカルによっても誘発され、酸素ストレスにより反応することが示唆された。Wgp7を有するトランスジェニック植物を作成し、これらが酸素あるいは二酸化イオウ、二酸化窒素ストレスに反応するかどうか検討中である。さらに誘発に必要なプロモーター領域の解析のためpWgp7-GUS遺伝子の組み換え体を作成中である。得られたタンパクを用いて抗体を作りWgp7産物の局在化を調べる。
