| Research Abstract |
先に提出した科研費交付申請書の、本年度の実施計画の3項目のうち、2項目は達成し得た。すなわち、正常表皮角化細胞と外毛根鞘細胞を手術標本より分離、培養し、培養細胞よりmRNAを抽出しWilks Primerを用いてRT-PCRを行い、1回目のPCR産物の電気泳動のゲル上より帯状のバンドを切り出し、そこからQIAGENのキットを用いてDNAを抽出し、それを鋳型として、nested primerを用いて2回目のPCRを行った。もう一度その産物の電気泳動のゲルより産物のDNAを回収し、PCR vectorとligationを行った。更に今後ligationの産物を大腸菌にtransfectし、cloningしてinsertのtyrosine kinaseのsequencingを行う必要がある。
RT-PCRの皮膚疾患への応用としては、他にアトピー性皮膚炎のダニ抗原誘発皮疹におけるTH_2系サイトカイン(IL4、IL5)およびTNFαのup-regalationを検出し、Journal of Allergy and Clinical Immunology誌にacceptされた。
