Research Abstract

活性酸素種は細胞内DNA損傷や細胞膜の酸化等を起こし、細胞の形態、生存、機能に致命的な問題をもたらす。このような活性酸素媒介の細胞損傷を防止するための方法として、抗酸化剤の摂取を介して人体め抵抗力を向上させることが知られている。ポリフェノールの主成分であるエピガロカテキンガレート(EGCG)は抗酸化、抗炎症、抗腫瘍また抗菌効果など様々な薬理作用を持つことでよく知られている。これまでの研究は、抗酸化剤としてのEGCGの抗癌効果について精力的に進められてきているが、本研究ではEGCGの様々な生理活性機能性に着目し、EGCGの生理活性が維持できる生体内分解吸収性高分子との複合体を調製し、再生医療のためのスキャホールドとして、生体組織再生及びその再生組織の保存技術への応用を試みた。まず、EGCGとラクチドーカプロラクトン共重合体(PLCL)をコーティング、抱接などの方法でEGCG/PLCLの複合体を作製し、得られたEGCG/PLCL複合体に対して表面特性(SEM)、熱的性質(DSC)、親水化程度(DCA)、官能期分布(FT-IR)、EGCG徐放性、及び力学的特性(引張強度、弾性率)等を測定した。さらに細胞毒性などの生体適合性を評価し、複合体に対する細胞の反応(付着、増殖、週期、移動、壊死など)と直接関連している細胞膜上の受容体との相互作用、細胞内シグナル伝達経路のみでなく、MMP-2活性、AktとNF-κB発現及びMAPK燐酸化を解明しEGCG/PLCL複合体の可能な作用機構を明らかにした。その結果、EGCG/PLCL複合体は再生医学・工学の分野に於ける人工血管、再狭窄防止ステント、創傷治療や、更にアトピー治療の薬剤の開発に応用可能だけでなく、哺乳類細胞と生体組織の非冷凍保存技術にも有効に応用できる知見が得られた。
