Research Abstract

平成20年度においては、開発ツールの1つである光合成生物の色素合成変異体を作製し、生体内でクロロゾームライブラリーを作製することに成功した。まずは、クロロフィルの20位メチル基転移酵素BchUの変異体を生体内で発現する光合成細菌の株を作製した。このことによって、バクテリオクロロフィル(BChl)cとdが変異体内で混ざって合成され、BChl dを基準とすると,0、20、30、60、80、100%の割合で持つクロロゾームが生体内でつくられていることが分かった。このように、1つの種を用いて、異なる割合でBChl cとdを持つ変異体が作製されたのは今回が初めてである。これは、当初目的としていた、変異体作製による有機合成過程の出発物質の多様化を満たしただけではなく、クロロゾームライブラリーを生体内で作製したことになる。このクロロゾームライブラリーの物性を調べるために、代表者は渡米し、ミズーリ州・ワシントン大学(セントルイス)のBlankenship教授のもとで、分光学的研究を行った。低温・常温における可視吸収スペクトルおよび蛍光発光スペクトル測定、または、CDスペクトル測定を行うことで、BChl cは、BChl aへのエネルギー伝達効率がBChl dよりも良いが、常温においては、連続する更なるエネルギー受容体がないと、光エネルギーのバックトランスファーが起こってしまうことが明らかとなった。
