Research Abstract

細胞の増殖と癌化の機構には、細胞内シグナル伝達のネットワークを構成するリン酸化酵素・脱リン酸化酵素・膜リン脂質・アダプター蛋白質等の分子群が重要な役割を担っている。これら分子群の細胞増殖・癌化に果たす役割を解析し、以下の諸点を明らかにした。1.細胞質分裂期の中間径フィラメントの嬢細胞への分配にsmall GTP結合蛋白質Rhoの標的分子の1つであるRhoキナーゼが必須であることを証明した(稲垣)。2.上皮細胞の分化に特異的なCキナーゼη分子種のゲノムを解析し、この遺伝子がマウス染色体12番長腕に位置することを明らかにした。またキナーゼATP結合部位エクソンを消失したマウス胚性幹細胞を得た(千田)。3.MAPKホスファターゼ(MKP)のmRNAレベル肝/肝癌系で解析した。MKP-2mRNAは、正常肝・再生肝・初代肝細胞では検出されず、原発肝癌および腹水肝癌で検出され、プログレッションとともに増大することがわかった。(菊池)4.ホスホリパーゼD(PLD)の活性化を抑制すると増殖刺激によるc-fos、c-junの発現が抑制されること、またアポトーシスに伴いPLD1のmRNAが著しく減少することを明らかにした。(中島)。5.PP2CはJNKシステムには影響を与えないが、p38システムを抑制すること、および、その作用点がp38を活性化するキナーゼであるMKK3やMKK6か、それらより上流にあることが判明した(田村)。6.細胞内情報伝達に関する不飽和脂肪酸を有する天然型ならびに類縁イノシトールリン脂質類、PI(45)P2、PI(3,4)P2、PI(3,4,5)P3の化学合成に成功した(渡辺)。7.アダプター型癌遺伝子産物Crkの主たるエフェクター分子C3Gが、Crkの発現により細胞膜へ移行し、酵素活性が上昇することを見出した。(松田)。8.ヒト・ミオシンホスファターゼ調節サブユニット(MYPT)の新規アイソフォームのクローニング、組織発現および遺伝子座の解析を行った。本分子は従来のMYPTとは異なり、心筋、脳特異的に発現しており、その遺伝子座は家族性拡張型および肥大型心筋症の原因と考えられる遺伝子と同じ領域に位置することが明らかとなった(中野)。
