Research Abstract

1.pRL1aペプチドのL^d分子結合アミノ酸残基とエピトープ残基
BALB/cマウス白血病RL♂1の細胞傷害性T細胞(CTL)認識拒絶抗原pRL1aペプチドのH-2クラスIL^d分子への結合に関与するアミノ酸残基およびCTLにより認識されるエピトープアミノ酸残基を検討した。pRL1aペプチドの各アミノ酸をそれぞれAlaで置換しアラニン置換ペプチドを合成し、L^d分子への結合を競合阻止によって検討したところ、8番目(P8)のLeuが主に結合に関与し、2番目(P2)のProが一部結合に関与していることが明らかになった。さらに、アラニン置換ペプチドをP815に感作し、pRL1a特異的CTLによる認識を検討した結果、pRL1aペプチド特異的ではあっても、pRL1a内で認識するアミノ酸残基のセットが異なることが明らかになった。RT-PCRによるCTLのVβ遺伝子の解析でも、用いられるVβは異なっていた。このことから、エピトープペプチドを認識するCTLに多様性があることが明かになった。
2.pRL1aペプチドによる抗腫瘍効果
pRL1aペプチドによる抗腫瘍効果を検討するため、Lysコアに8価のpRL1aを結合させた多価pRL1aMAP(multiple antigen peptide)を合成してBALB/cマウスに投与し、CTLの生成と腫瘍増殖に及ぼす効果を検討した。その結果、pRL1aペプチドそれ自身では効果がないが、pRL1a MAPによる免疫でBALB/cマウス脾細胞にpRL1a特異的CTLを誘導した。さらに、in vivo感作の際に、CD4^+T細胞とAPCが必要であることを明らかにした。また、pRL1a MAPによるin vivo感作でRL♂1腫瘍の増殖を特異的に抑制できることを明らかにした。
