Research Abstract

一部の腫瘍細胞や固型腫瘍において恒常的に一酸化窒素合成酵素(NO synthae,NOS)が発現しNOを産生していることが知られている。このNOSはチトクロームP-450還元酵素に類似していることが知られており、本年はこのNOSに焦点を当てて、制癌剤感受性増強効果および固型腫瘍のNO産生動態とその意義について検討した。
(1)固型腫瘍局所にNO放出剤を投与して腫瘍血管の透過性を亢進させることにより、高分子制癌剤であるSMANCSのdrug delivery system (DDS)改善傾向を認めた。(2)またNOSの基質であるL-arginineの投与により固型腫瘍局所における内因性のNO産生を高める事により、腫瘍血管の透過性を更に亢進させることが明らかになった。(3)AH136B固型腫瘍のNO産生動態をelectro spin resonance (ESR)を使用して検討したところ、比較的多量のNO産生を認めた。(4)さらに増殖の速い腫瘍ほどNO産生量も増加する傾向を認めた。
