Research Abstract

本研究は、ATP駆動型トランスポータとチャネルの構造的・機能的相違と機能的な協関性を明らかにすることを目的として行った。その結果、
1)トランスポーターであるMDR1とATP感受性KチャネルのサブユニットであるSUR1は、どちらもABCスーパーファミリーに属するが、ヌクレオチドとの相互作用が大きく異なることを明らかにした。(植田)
2)SURとKirのそれぞれのサブタイプのcDNAを単離し、培養細胞に発現させ、電気生理学的に解析した。その結果、心筋のATP感受性KチャネルがSUR2AとKir6.2の組み合わせによって、血管平滑筋のATP感受性KチャネルがSUR2BとKir6.1の組み合わせで構成されていることが示唆された。(倉智)
3)実験虚血条件下、急性単離した海馬ニューロンにおいて、細胞内ATPの減少、それに続くCa濃度の上昇が見られた。細胞内Caは、GABA_A受容体の機能には直接影響しないが、ATPは直接的に作用することを証明した。(赤池)
4)Na-Kポンプの特異的阻害剤であるウアバインの結合部位とH^+,K^+-ATPaseの阻害剤であるSCH28080結合部位は、それぞれの蛋白の異なる領域であることを明らかにした。(竹口)
5)グルタチオン合成阻害剤とMRP阻害剤を併用することによって、癌細胞の多剤耐性を効果的に克服できることを明らかにした。(秋山)
