Research Abstract

MRPの抗ペプチド抗体で免疫組織化学的に、気管支線毛細胞、副腎皮質、腎尿細管などに高く、消化管で低く、神経系にはないなど正常組織の発現を検討した。
臨床例の肺、大腸、胃の癌と正常部でMRPのmRNAと蛋白質の発現を見た。肺扁平上皮癌では9例中8例で高く、4例は抗癌剤感受性株の3.6倍以上で、肺扁平上皮癌の耐性にMRPの関与が強く示唆された。
MRP高発現細胞株C-A120とシスプラチン耐性株KCP-4のmembrane vesicleを用いて、ロイコトルエンC4(LTC4)の輸送とその輸送の薬剤での阻害について検討し、二つの細胞のグルタチオン抱合体ポンプを比較をした。両細胞のmembrane vesicleはともにin vitroでLTC4を能動輸送し、Km値は1.550、0.455μMでVmaxは308、4.447mol/ng/proteinであった。C-A120ではLTD4でKCP-4ではグルタチオン抱合体シスプラチンでより強くLTC4の輸送が阻害された。MRPはグルタチオン抱合体排出ポンプとして働く事が明らかになった。また、KCP-4細胞のグルタチオン抱合体排出ポンプはMRPとは特異性が異なる事が示唆された。
グルタチオン合成阻害剤BSOはC-A120のmembrane vesicleのLTC4の輸送を阻害せず、ビンクリスチンの細胞内濃度を上昇させ、耐性度を低下させた。BSOはグルタチオン濃度低下させて、耐性を克服したと考えられ、ビンクリスチンがグルタチオン抱合され輸送されている事が示唆された。
ピリジン誘導体PAK104PはP-糖蛋白質の機能を阻害するが、MRPのLTC4の輸送も阻害し、C-A120のビンクリスチン耐性を克服した。PAK104Pは広いスペクトラムの有望な抗ガン剤多剤耐性克服薬剤と考えられた。
