Research Abstract

本研究はエイズ病態の進行及びそれとは逆の発症を未然におわらせる諸条件をウイルス遺伝子及び遺伝子産物と宿主細胞との相互作用を軸に解明することを目的に開始された。本年度は、独自に開発した新規発現ベクター、センダイウイルスによるウイルス感染抑制、ケモカインSDF-1の大量生産が可能となり、SDF-1の結晶化とX線解析へと進んでいる。同じ系でHIV-1のエンベロープ糖蛋白の大量生産も可能となったので、SDF-1の受容体であり、HIV-1の細胞侵入に必須の副受容体CXCR4を軸としたウイルス・細胞相互作用とSDF-1による感染ブロックの機構の解明が加速されることが期待される。一方、マウスのCXCR4もヒトのそれと同様にHIV-1の副受容体として機能できることが明らかとなり、HIV-1の厳しい種特異性の決定に、少くともCXCR4は無関係であるという以外な事実を提示し、国際的に大きな影響を与えた。以上のケモカインとケモカイン受容体を中心とする活発な研究に加えて、潜伏感染細胞からのHIV-1活性化における各種のアクセサリー遺伝子の役割、HIV-1感染における、新たな細胞表面分子FRP(CD98)の関与の可能性、長期未発症者におけるNef蛋白特定エピトープに対する高い抗体応答など新しい展開もあった。また、感染者個体内のウイルス変異の解析がさらに進み、比較的緩慢な経過をとる症例にエンベロープ糖蛋白のV2領域の重複が高率に出現するなどの新たな知見が得られた。
