Research Abstract |
We have analyzed phytplasma in terms of molecular evolutionism, and studied in order to establish systems of genome analysis and molecular diagnosis. First, we gathered various separated roots of phytoplasma and refined the genome by the CTAB method. Then we amplified 16SrRNA genes by PCR and determined the whole base arrangemant by direct sequences. Further, we refered the newest base data of 16SrRNA genes in microbes including phytoplasma in other countries, using data base of the large computer, and compared the results to the base arrangement data we had before. Therefore, We have constructed the molecular genealogical family tree by large analysis program "ODEN" and proceeded "Bootstrap" to analyze reliablility. Under the analyzed data, we have established the classifying system of phytoplasma. Further, we have applyed the classification of "species" to phytoplasma by the above data and system, designed and composed the unique primer. Also we have tried to classify the unknown separated roots of phytoplasma in the various area of Asia.
