Research Abstract |
The mechanism of aging process was investigated by research on stress response during aging and the effect of orally hormons on the response by use of senescence-accelerated mice (SAMP1 // Fky and SAMR1 / Fky). The results obtained and the view advanced are as folows ; 1)The steady-state levels of HSP70 expression and oxidized protein in liver and brain from both strains of SAMs gradually increased with age. The steady-state levels of HSP70 expression was a positive linear function of oxidized protein levels. These results suggest that stresses accumulate in vivo in the course of aging. Thus, senescence can be referred to as a kind of stresses. 2)HO-1 was not detected throughout lifespan of both strains of SAMs. HO-2 in 12-month-old SAMR1 / Fky and 7-month-old SAMP1 // Fky increased significantly by 2-times and 1.5-times respectively when compared with that in 2-month-old mice in either strain and subsequently decreased, whereas HSP70 in both strains of SAMs gradually increased with age.

Between HO-2 expression and iron concentration there was a positive correlation. These results indicate expression of HO-2 that is a constitutive type depends on contents of iron during the process of aging. 3)Induction of HO-1 and HSP70 in response to paraquat administration significantly decreased with age in both strains of SAMs, although oxidized protein levels gradually increased with age. Moreover, oxidized protein levels were suppressed when HO-1 was induced, then, remarkably increased after decreases of HO-1 induction. This result suggest induced HO-1 functions as an antioxidant at early phase and a prooxidant at later phase. 4)Neither melatonin nor DHEA administration changed the basal levels of HSP70 and HO-2 expression compared to control diet. However, inductions of HSP70 and HO-1 expression by paraquat administration were significantly higher in SAMP1 // Fky and SAMR1 / Fky administered melatonin compared to control mice, respectively. The induction of HO-1 expression were significantly higher in SAMP1 // Fky administered DHEA compared to control mice. These data indicate that orally melatonin and DHEA administration affect a tolerance for oxidative stress. Less
