Research Abstract |
Effects of restraint stress on ET-1 and bNOS mRNA expression in the brian were investigated in normotensive rats using RT-PCR and RNAase protection assay.Although expression of ET-1 mRNA was highest in the brain stem, that in the brain cortex and hypothalamus showed a significant decrease in response to the stress. While bNOS mRNA expression was highest in the cerebellum, that in the hypothalamus showed a significant increase in response to the stress. Since ET-1 and bNOS gene expression was decreased and increased, respectively, in the hypothalamus under acute stress, it was suggested that ET and NO in the brain also mediate stress mechanism.^ハ In oeder to elucidate the role of NO in the central regulation of blood pressure, effects of i.c.v administration of NOC-18 as NO donor and i.v.administration of L-NAME on c-fos gene expression ware determined by in situ hybridization in the brain of normotensive rats. On the contrary, L-NAME resulted in a significant increase in blood pressure and c-fos mRNA in the PVN and NTS.These results suggested that NO plays an important role in the central depressor mechanism. Effects of restraint stress on nuclear mRNA expression of oncogenes, CRF,and CRF receptor were studied in the hypothalamus of SHR-SP.After 2 hrs of restraint stress, CRF and CRF receptor mRNA expression were increased in SHR-SP,suggesting a hyperreactivity of the pituitary-adrenal axis to the stress in SHR-SP.mRNA expression of CRF hnRN,Ac-fos, jun B,and NGFI-B in the PVN was increased after 30 min of stress in SHR-SP.The findings were also seen in rats before development of hypertension, suggesting the increased gene expression of CRF and oncogenes is not the results of hypertension but is related to the pathogenesis of hypertension. All these results suggest that various vasoactive substances in the central nervous system could have key roles in the pathogenesis of stress and stress-induced hypertension. Less
