Research Abstract |
OBJECTIVES : Recent studies revealed that bFGF administered through arteries had a significant angiogenic effects. However, it has not been clarified whether bFGF directly injected into the myocardium has a similar effects of angiogenesis or not. The purpose of this study is to investigate the angiogenic effect of bFGF locally administered to the ischemic myocardirm. MITHODAS : 1) We made a model of ischemic myocardium by ligating the left anterior descending artery in mongrel dogs. We directly injected human recombinant bFGF into the myocardium or left one on the epicardium. We examined the grade of angiogenesis by coronary angiography and pathohistological examination 3 to 4 weeks after surgery. We administered bFGF under following condition ; (1) 0.9% saline, (2) bFGF 10mug, (3) bFGF 10mug + heparin sulfate 1mg, (4) bFGF 30mug, (5) bFGF 30mug + heparin sulfate 1mg, (6) bFGF 300mug + heparin sulfate 10mg, 2) We administered bFGF 300mug + heparin sulfate 10mg to a myocardial ischemia model of Japanese monkeys. 3) We administered bFGF 300mug + heparin sulfate 10mg to a myocardial ischemia model of mongrel dogs after performing coronary artery bypass from the left internal thoracic artery to the left anterior descending artery. RESULTS : We could find collateral circulation to a certain extent around the infarcted area, however it was not significantly promoted by bFGF in experiments 1), 2), or3). COMMENT : After experiments of 1) (1) - (5), We thought that larger doses might be required to have angiogenic effect. However, large amount of doses up to 300mug could not express a significant effect. Then we suspected the difference between species, and we performed similar experiments in Japanese monkeys. But even in Japanese monkeys model, significant increase of angiogenesis could not recognized. Now we suppose that bFGF did not have significant effect of angiogenesis because it may be inactivated before uptake into the endothelial cells or myocardium.
