Research Abstract

In the society of advanced age, the osteoporosis and periodontitis are important factors to keep QOL (quality of life).
The purpose of this research is to clear up the relevancy between osteoporosis and periodontal tissue (especially cementum) through the studies of contact microradiography, cell culture of cementoblast and extraction of cementocyte DNA from paraffin specimen.
(1) Contact microradiograpy : Osteoporosis causes a fracture of thighbone. The reason of this fracture is the excess absorption of calcium and phosphte of bone by osteoclast. Though a mandibular bone is the same type bone (tubular bone) as thighbone, the clear effect from osteo-porosis to mandibular epecially alveolar bone could't be observed.
(2) Cell culture of cementoblast : The determinaton of cementoblast is based on the location that the cell attached to the superficial surface of root. But it is very difficult to determine the cementoblast accurately. So we would like to establish the determination of cementoblast using histochemistry and flowcytemetry marked vitamin D3 receptor. According to histochemical study, we could observe the fluorescence reaction of vitamin D3 receptor on the surface of cementoblast. But we could not collect the cementoblast using flowcytemetry and cell culture.
(3) Extraction of cementocyte DNA from paraffin specimen : To extract the cementocyte DNA,we used DNA isolator kit (Wako Co.). After that we tried to detecat beta actin, osteopontin and osteonectin using PCR method. We could find those in osteocyte specimen, but we could't observe those in cementocyte specimen clearly. Hereafter we must continue molecular research to establish the determination of cementoblast, especially we should pay attention to 47kbp protein.
