Research Abstract

本研究の目的は、申請者らが発見したインスリン産生細胞の新しい増殖因子Reg蛋白の作用機序を明らかにし、組換えReg蛋白を用いてインスリン産生細胞を再生・増殖させるという糖尿病治療法開発への道を開くことである。
平成8年度の研究は、当初計画通り順調に進行し、以下の成果を得た。
1.ES細胞染色体上での相同組換えの手法を用いてReg遺伝子欠損マウスを作製することに成功した。
(1)染色体上の遺伝子が変異Reg遺伝子と入れ換わったES細胞株をマウス胞胚に注入して代理母に移入したキメラ個体を得た。
(2)(1)で得られたキメラマウスの交配を行ない、Reg遺伝子欠損マウスを得た。
2.ヒト及びマウスのReg遺伝子ファミリーの遺伝子座を決定した。
(1)ヒトREG遺伝子ファミリー(REGIα,REGIβ,HIP/PAP,RS)の遺伝子座の決定をFISH法およびYACクローンの解析により行ったところ、当該遺伝子ファミリーが全て第2番染色体p12に存在し、セントロメア側からHIP/PAP-RS-REGIα-REGIβの順序で並んでいることが明らかとなった。
(2)マウスReg遺伝子ファミリー(RegI,RegII,RegIIIα,RegIIIβ,RegIIIγ)の遺伝子座をFISH法により解析したところ、当該遺伝子ファミリーが全て第6番染色体上の極めて近接した領域に存在していることが明らかになった。
3.平成7年度に作製したReg蛋白過剰発現マウスの解析を行った。
Reg蛋白過剰発現マウスの膵β細胞の形態、大きさは正常マウスとほとんど変わらなかったが、Reg蛋白過剰発現マウスから分離した膵ランゲルハンス島は正常マウス膵ランゲルハンス島と比較して15〜3倍のDNA合成を示し、Reg蛋白の過剰発現によりβ細胞の増殖能が上昇することが明らかにされた。
