Research Abstract |
To understand the process of speciation, organization of nervous system and mechanism of sex-determination, I have analyzed mating behavior mutations in Drosophila melanogaster. 4000 P-element tagged lines were constructed and sterile or semi-sterile stocks were selected from them.Each sterile strain was tested for its mating behavior, because files with severe mating behavioral defects look like sterile. I obtained several behavior mutation candiates out of 140 sterile strains. Females of the strain #933 have abnormal genitalia and surprisingly they try to court with males but females. More detailed analysis showed that this mutation was a hypomorphic allele of transformer-2. Three mutant stocks with visible phenotype as well as courtship behavior defect in males were identified as new alleles of previously characterized genes. One such mutation was an allele of anterior-open (aop) and this had rough eyes and depigmented tergites. Other two stockes are alleles of scabrous (sca) and rotaed-abdoment (rt). Using males of a D.simulans stock, 37 female mating behavior mutation candidate were obtained from above P-tagged lines. Each mutant showed high repeatability for their mating tests.
