Research Abstract |
Rat megakaryocytes (MK) were isolated by an immunomagnetic beads method as described in the year report of 1994, and cultured MK and PPF were observed electron microscopically. 1)PPF assay of thrombopoietin activity : a)as for the stromal cell enhancement effect on PPF activity, TPO was compared to IL-6 and Epo using coculture of porcine endothelial cells (PEC) and rat MK.Varying concentrations of TPO (lpg/ml-10ng/ml) showed a greatly enhanced PPF on the coculture system like IL-6 and Epo. b)as for synergism of hemopoietic factors effect on PPF,there was no synergism between TPO and IL-6 or Epo. Similarly, IL-3 was not synergistic to TPO as well as IL-6, but synergistic to EPO. 2)Biological analysis of stromal cell enhancement effect on PPF was not advanced because of mycoplasma infection of stromal cells such as KD cells. 3)When MK and PPF were observed electron microscopically, there was no significant difference inthe ultrastructure between MK treated with TPO and those treated with IL-6 or Epo, while TPO tends to increase more demarcation membrane complex (DMC) than IL-6 or Epo. PPF is thought to be derived from dissciation of DMC and thread like structure of PPF might be due to incomplete development of DMC because of in vitro culture system.
