Research Abstract

Chloramphenicol acetyltransferase (CAT) transgenic mice were used for study the variation of intron enhancer (Emu) activity during the late stage of B cell development. Science the expression of CAT transgene was regulated by VH promoter and Emu, a change in the CAT activity provides a measure of transcriptional activity of Ig genes. When the resting B cells were stimulated by cross-linking with goat anti-IgM antibodies, the CAT activity decreased approximately 1/10. On the other hand, 20-30% increase in the CAT activity was observed by stimulation with lipopolysacharide (LPS). The stimulation of B cells with LPS caused an increase in the CD43^+B cells that possesed a higher CAT activity than CD43-B cells. On the other hand, CD43^+PNA^+B cells obtained by stimulation with antigens in vivo also showed increased CAT activity, suggesting the enhanced transcriptional activity of Emu. These results suggest that Emu activity was augmented in B cwlls where somatic mutation is induced with high frequency.
