Research Abstract

In order to evaluate the individual susceptibility to mutagen, we have measured spontaneous or mutagen(X-ray or colchicine)-induced micronucleus frequency in human lymphocytes from a normal healthy population, and have analyzed statistically for interindividual variance. As a result, there was a significant difference in the mutagen-induced micronucleus frequency between individuals and this was indicative of interindividual variance in susceptibility to these mutagens. Then, we have investigated the relationship between these micronucleus levels and serum antioxidative activity or antioxidant micronutrients(vitaminsB12, CandE,beta-carotene and folic acid) levels. Serum antioxidative activity measured by the electron spin resonance spectrometry (ESR) was strongly correlated with the serum levels of water-soluble micronutrients such as Vitamin C and folic acid. Kinetochore-labelling study revealed that folic acid and vitamin C are major variables for induction of micronuclei in lymphocytes. Serum folic acid level was negatively correlated with the kinetochore-positive and-negative micronucleus frequency and vitamin C was positively related to the formation of kinetochore-positive micronuclei. These reults suggest that interindividual variance in chromosomal response to mutagens(X-ray and colchicine) in normal population may be a real phenomenon, and that individual susceptibilities to these mutagens are unrelated. Our data also suggest that the baseline micronucleus level which is strongly associated with the levels of certain serum micronutrients(folic acid and vitamin C) is not of predictive value for the susceptibilities to these mutagens.
