Research Abstract |
The purpose of this study is to elucidate the distribution of blood group A and B determinants by isolating blood group A,B,H-active polyglycosyl peptides from human blood group AB gastric mucosa. Blood group A,B,H-active glycopeptides were extracted from human blood group AB gastric mucosa by the phenol/ethanol method after pepsin digestion. The glcopeptides extracted were chromatographed on an anti-A active immunoadsorbent column. The eluate from the anti-A column was successively chromatographed on an anti-B active immunoadsorbent column. The eluate from the anti-B column was found to possess both A and B activity of equal magnitude by hemagglutination inhibition tests. Then, the glycopeptides were subjected to pronase digestion. The resultant product was split into several fractions according to the molecular weight of the digested glycopeptides by ultrafiltration. A fraction (100-50kD) possessing both A and B activity was chromatographed on an anti-A column. The eluate from the anti-A column was successively chromatographed on an anti-B column. The eluate from the anti-B column was shown to possess both A and B activity of the same magnitude and not to possess H activity by hemagglutination inhibiton tests. The evidence obtained from these experiments strongly indicates that in the blood group specific substances from human blood group AB gastric mucosa, a certain proportion, if not all, of the carbohydrate chains showing blood group-specificity possess both A and B determinants.
